Effects of nuclear factorκB activation on expression of inflammatory mediators of alveolar macrophages associated with acute necrotizing pancreatitis

Objective To investigate the effects of nuclear factorκB (NF-κB) activation on expression of inflammatory mediators of alveolar macrophages (AMs) associated with acute necrotizing pancreatitis (ANP) and lung injury. Methods Thirty adult SD rats were randomized into 5 groups (n=6) of the normal control group, 1-h group, 3-h group, 6-h group and 12-h group after ANP induction. ANP was induced by intraductal administration of 5% sodium taurocholate, while the normal control rats received an infusion of normal saline. AMs were harvested by bronchoalveolar lavage. The protein content of bronchoalveolar lavage fluid (BALF), the myelopOeroxidase (MPO) in lung tissue, and TNFαand NO secreted by AMs were determined. The expression of TNFαmRNA and inducible nitric oxide synthase (iNOS) was detected. The histological changes in lung was scored in a blinded fashion and the expression of NF-κB was determined with immunohistochemistry. Results The activation of NF-κB was seldom seen in AMs of normal lung tissue, while it could be observed in ANP groups. NF-κB moved from cytoplasm into nuclei with time prolongation. The lung injury was gradually aggravated with disease progression. The level of MPO of the lung tissue and protein content of BALF showed a steadily significant increase with time prolongation. TNFαand NO secreted by AMs were gradually elevated and peaked at the 6th h but decreased at the 12th h. The change in expression of TNFαmRNA and iNOS mRNA was similar to that in TNFαand NO. There were significant differences in all the parameters between the control group and ANP groups (P0. 05). Conclusions NF-κB is activated after the onset of ANP to upregulate inflammatory mediators secreted by AMs to aggravate lung injury.
