High level expression of recombinant VEGF in Escherichia Coli

He Yanzheng, Zhang Peihua, Xiao Gutian, et al. Department of Surgery, No. 9 People's Hospital, The Second Medical University of Shanghai, Shanghai 200011
To study its structure and function, a high level expression plasmid, PRL621/VEGF, which yielded about 30% of total cell protein of Escherichia Coli, was constructed. The inclusion bodies which the uroduction stayed inside were isolated, denatured and then initially renatured. Chorioallantoic membrane (CAM) assay showed the renatured VEGF promoted new vessel formation. Its first 15 N-terminal amino acid sequence was identical to that of the natural protein.
