Dynamic observation of pathological change and activation of nuclear factor-kappa B in the intestinal mucosa of rats after diffuse brain injury

Objective To explore the effect on the intestinal mucosal barrier dysfuction of rats after diffuse brain injury by studying the dynamic changes of NF-κB expression and apoptosis in the intestinal mucosal endothelial cell.Methods The animal′s models established by Marmarou were used to created diffuse brain injury.150 male Wistar rats were randomly divided into 9 groups according to the time after injury(1,2,4,8,12,24,48,72,168 hours group) and 1 control group.Expression of NF-κB and the number of intestinal epithelial apoptotic cells between control and injury groups was observed by using the immunohistochemical staining and the TUNEL methods.Results The optical density of NF-κB of injury groups was much higher than that of control group.The number of apoptotic cells began to increase at 1 h,peaked at 12 h and decreased gradually,which was significantly increased in injury groups compared with control group during whole observation period(P0.01).Conclusion NF-κB activated by cellular stress,and apoptosis of intestinal epithelial cells might play an important role in the development of intestinal mucosal barrier dysfunction after diffuse brain injury.
