Determination of natamycin in fermentation broth by RP-HPLC

Objective An RP-HPLC method for the determination of natamycin in fermentation broth was developed. Methods The fermentation broth was extracted by methanol and the extraction was analysed by HPLC. The sample was separated at room temperature on a Hypersil BDS C_~18 column (5μm, 4.6mm×200mm) eluted at a flow rate of 1.00 ml/min with a mobile phase of methanol-water-acetic acid (60∶40∶5, V/V/). Natamycin was detected and quantified by the absorbance at 302nm. Results The intra-day RSD and inter-day RSD were 0.15% (n=6) and 0.90% (n=6), respectively. The mean recoveries were 99.84%, 101.18% and 101.41% versus the added authentic sample at the concentrations of 92, 184 and 368μg/ml, ~respectively . Conclusion This method is simple, accurate, highly specific and reliable for the determination of natamycin.
