The studies on the genetic engineered strain producing 5-O-avermectins

Using the strategy of gene insert disruption, a 1.5 kb apramycin resistance gene fragment was inserted into the SalI site within the aveF gene,As a result, the recombinant disruption plasmid Ppc-5 was constructed. The plasmid pPC-5 was transferred into the Streptomyces avermitilis D2-14 to obtain recombinant strain AV-F1. The products of strain AV-F1 by HPLC showed mainly two components, which were 5-O-avermectin 1a and 5-O-avermectin 2a, respectively. Moreover, it produced a small amount of avermectin B1a. The recombinant 5-O-avermectins producing strain AV-F1 could provide available material for chemical synthesis of new avermectins derivatives.
