Effect of enalapril on angiotensin converting enzyme and left ventricular hypert rophy in the spontaneous hypertensive rats

Objective To investigate the influence o f angiotensin converting enzyme (ACE) activity and enalapril on the left ventricul ar hypertrophy (LVH) in the spontaneous hypertensive rats(SHR).Meth ods 40 12-week- old SHRs were divided randomly into enalapril 3 0 mg/(kg·d) treatment group and control group, at the same time, WKY with the match age was normal control. The thickness of the wall of left ventricular, the BM/LV, CVF and PVCA were measured by pathological examination and morphological analysis. The concentration of AngⅡ was measured with RIA. The activity of ACE was determined wit fluorescence analysis. Results (1) BM / LV in SHR-T was similar to that of WKY-C (P0.05). (2)The concentrati on of Ang Ⅱin heart of SHR-C (5.780±3.734 ng/mg tissue) was much higher than that of WKY-C (2.723±0.84 ng/mg tissue). But after the treatment of enalapril, there was no significant difference of AngⅡconcentration between SHR-T (2.75 7±2.717 ng/mg tissue) and WKY-C. (3)The activity of ACE in SHR-T, SHR-C and WKY-C were 0.47±0.12 units/〔ml·mg(tissue)〕, 0.60±0.13 units/〔ml·mg(tissu e)〕, 0.45±0.19 units/〔ml·mg(tissue)〕, respectively. The activity of ACE in heart of SHR-C was higher than the other groups (P0.01), and there is no d ifference between SHR-T and WKY-C (P05). (4) CVF and PVCA in heart of S HR-T were 1.98%±1.57% and 0.68%±0.19%, respectively, which were lower than th ose of SHR-C 〔(5.11±2.25)% and (1.20±0.19)%, P0.05)and similar to those of WKY-C (1.70%± 0.63% and 0.5%±70.17%, P0.05). Co nclusions ACEI can inhibit the activity of local ACE, decrease the concentration of AngⅡ and reversed LVH.
