Study on protoplast preparation and regeneration of Morchella crassipes

The protoplast formation and regeneration of Morchella crassipes were studied. The optimum conditions for protoplast formation were determined. In details, the enzymolysis system consisted of 4% snailase, 5% lywallzyme and 4% ellulase. And 0.6 mol/L KCl phosphate buffer (pH 6.98) was used. The enzymolysis was conducted at 30℃ for 4 h. The age of cells for protoplast preparation was 56 h. The protoplast yield reached 5.3×104 cells/mg. On the PSA (potato sucrose agar) medium contenting 0.3 mol/L KCl and 0.3 mol/L inositol, the regeneration rate was 9%.
