Study on the Detection Methods of Long-term Storage Pollen Viability of Paeonia lactiflora Pall

The viability of Chinese peonies varieties pollen preserved in different temperature(room temperature,4 ℃,-20 ℃)and storage time were detected by I2-KI coloration and TTC coloration,and the germination rate of pollen were detected at the same time by germination in culture medium,to research the correlation of I2-KI,TTC and cultured in vitro.The results were as follow:the pollen viability and germination rate of Chinese peonies varieties reduced gradually as the storage time,and the descent of germination rate was quickest at room temperature;the germination rate of pollen preserved at room temperature for 7 days or 4 ℃ for 6 months reduced to zero,and the germination rate of pollen preserved at-20 ℃ for 9 months reduced to 1.96%.The correlation coefficient between I2-KI,TTC with germination in culture medium,respectively were extremely significant or significant correlate.Compared with TTC,the results of I2-KI in 4 ℃ preserved for 4 months and-20 ℃ preserved for 7 months were more accurate.
