Astrocyte dysfunction and central nervous system diseases

Astrocytes, the most abundant cells in the central nervous system(CNS), are a critically structural and functional part of the tripartite synapses and the neurovascular unit. They are also closely related to other types of cells in the brain. Astrocytes respond to virtually all pathological conditions in the CNS such as infection, trauma, stroke and neurodegenerative diseases by a process commonly referred to as reactive astrocytes, which involve changes in morphological and functional heterogeneity of astrocytes. Indeed, recent studies suggest that the involvement of astrocytes in the etiology and progression of CNS diseases is likely caused by the loss of their normal homeostatic functions and/or the gain of toxic functions. This review describes the multiple roles of astrocytes in the CNS diseases, discusses the diversity of astroglial responses in neurological disorders and argues that targeting astrocytes may represents an new therapeutic strategy for stroke, epilepsy, Alzheime's disease(AD) and Parkinson's disease(PD) as well as other CNS diseases.
