Determination of Rutin,Ginsenoside Rb_1 and Quercetin in Gynostemma pentapylla by HPLC with Programmed Wavelength UV Detection

Objective:To study and establish the quantitative analysis method for rutin,ginsenoside Rb1 and quercetin in Gynostemma pentapylla by HPLC with programmed wavelength UV detection.Method:HPLC was performed on TSK ODS-100V(4.6 mm×250 mm,5 μm),the mobile phase consisted of acetonitrile and water(containing 0.1% phosphoric acid) at a flow rate of 1.0 mL·min-1.The column temperature was set at 30 ℃,and the eluate was detected by programmed wavelength(0-14 rnin,257 nm;14-18 min,203 nm;18-30 min,257 nm).Result: Quantitative analysis of HPLC showed that the linear ranges of rutin,ginsenoside Rb1 and quercetin were 0.171 2-1.284 0 μg(r=0.999 8),0.828 0-6.210 0 μg(r=0.999 9),0.204 8-1.525 7 μg(r=0.999 8) respectively.The average recoveries of rutin,ginsenoside Rb1 and quercetin were 97.33%(RSD=2.02%),98.61%(RSD=1.70%),98.38%(RSD=2.59%) respectively.Conclusion: The method set up is accurate,feasible and reliable,therefore available for the quality control of G.pentapylla.
