The influence of hyperglycemia on the neurons following traumatic brain injury.

Objective To explore the influence of hyperglycemia to the neurons and it,s mechanism following traumatic brain injury.Methods Adult male SD rats were randomly divided into 3 groups: normal control group,traumatic brain injury group and insulin treated group,the blood glucose concentrations of the rats were measured before and after injury.RT-PCR and western-blot were applied to detect the expression of GLUT-3 genes and the change of GLUT-3 protein,respectively.The NSE and NSE positive-Staining cells were examined using immunofluorescence staining.Results The blood glucose concentration increased markedly in traumatic brain injury group compared with controls(P0.05).The expression of GLUT-3 and cellular NSE increased in the injured cortical tissue in both traumatic brain injury group and insulin treated group(P0.05)compared with controls.The significant increase in expression of GLUT-3 and cellular NSE and decrease in NSE positive-Staining cells were observed in the insulin treated group at 12,24,48,and 72 h after injury,respectively(P0.05).Conclusions The hyperglycemia after traumatic brain injury increases neuron damage by decreasing the expression of GLUT-3.
