Determination of arsenic speciation in aquatic products by high performance liquid chromatography with atomic fluorescence spectrometry

Objective To establish a method to determinate arsenate,arsenite,monomethylarsonic acid and dimethylarsinic in aquatic products by high performance liquid chromatography( HPLC)- hydride generation( HG)- atomic fluorescence spectrophotometry( AFS). Methods The samples were hot- dipped in 90 ℃ in 0. 15 mol / L nitric acid solution for 2. 5 h,extracted,centrifuged,defatted,purified by C18 column and filtrated. Samples were isolated on PRPX- 100 negative ion exchange column by HPLC- AFS,with the mobile phase as phosphate buffer solution( p H = 5. 9),and the 4 arsenic species in the extract was detected and analyzed with retention time for qualification and the external standard method for quantification. Results Good linearity was obtained when the concentrations of arsenate,arsenite,monomethylarsonic acid and dimethylarsinic were within0 ng / L- 100 ng / L,with the correlation coeficient( r) ≥0. 995 0 and the detection limit within 0. 02 mg / kg- 0. 05 mg / kg.When the matrix spiked recovery samples were at 0. 2 mg / kg,0. 4 mg / kg,0. 8 mg / kg in grass carp,the recoveries of arsenate,arsenite,monomethylarsonic acid and dimethylarsinic were within 93. 5%- 105. 1%,71. 1%- 84. 1%,72. 1%- 107. 8%and 80. 2%- 90. 5%,respectively. Conclusion The method is simple,accurate and reliable with high applicability. It can be used to analyze arsenate,arsenite,monomethylarsonic acid and dimethylarsinic in aquatic products.
