In Vitro Selection of A High - Proline- Producing Variant from Rice Callus and Studies on Its Salt Tolerance

This paper reported the selectionof a high-proline-producing variant ohtamed by 6to 60Coγ-ray radiation followedby selection with hydroxy-L-proline(Hyp) from immature embryo callusculture of Oryza sativa L. cv.Shuangfeng No. 1. The Hyp concentration used for selection was 3. 0 mmol/L. The selected variant had a highertolerance to Hyp. The proline contentof the variant was 2. 6-folds as high asthe original type (Table 1 ). When exposed to a high NaCl concentration,both the increase of proline content(Fig. 3) and salt resistance (Fig. 2)in the variant were greater than in theoriginal type. This result indicated thatan increase of endogenous proline content does strengthen salt tolerance ofthe cultured cells in rice.The high-proline-producing variant contained more soluble soluble (Fig.4 ), Na+ and K+ (Fig. 5) than the original type when cultured on a medium with 100 ̄ 300 mmol/L of NaCl,and the K+/Na+ ratio was the same asthat of the original type. Water content of the variant decreased a little under NaCl stress (Fig. 6). All of thesecharacteristics indicated that the comoregulation in the variant wasstrengthened when exposed to saltstress (Table 3).
