An Experimental Study on ICAM-1 Expression of Pulmonary Microvascular Endothelial Cells Induced by LPS and Its Mechanism Through Nuclear Factor-kappa B Pathway

Objective To study ICAM-1 expression of pulmonary microvascular endothelial cells (PMVEC) induced by LPS and its mechanism.Methods To measure ICAM_1 expression (by immunocytochemistry) stimulated by 100ng/ml LPS at 0h, 2h, 4h, 6h, 8h or 10ng/ml, 50ng/ml, 100ng/ml LPS at 6h and the activation of NF_κB by EMSA. Those to be compared with which were inhibited by PDTC. Results The expression of ICAM_1 was highly dependent on the stimulation of LPS in manners of dose and time. The stimulation quickly resulted in NF_κB translocating to nuclei, which peaked at 1h and then went down. PDTC could significantly decrease the expression. Conclusions The induced activation of NF_KB by LPS starts up ICAM_1 expression.
