Aim: To study the protective and therapeutic effects of paeoniflorin to rat's hepato-cellular damage in vitro. Methods: Primary culture damaged hepatocytes induced by D-galac-tosamine and/or endotoxin. The protective and therapeutic effects of paeoniflorin were observed by detecting the changes of cytomorphology and biochemical criterion such as lactic dehydrogenase (LDH). glutamic oxaloacetic transaminase (GOT), superoxide dismutase (SOD). Result: The structures of cell surface, caryon, endoplasmic reticulum , Golgi complex, mitochondrion , lysosome returned to normal in different protective and therapeutic groups which were corresponding to different damaged one. The expressions of LDH, GOT, SOD in different protective and therapeutic groups were obviously lower than those in damaged one (P0.05-0.01). Conclusion: paeoniflorin can in(?)rease the stability of hepatocyte and lysosome membrane, so that it has the effects for protecting and treating hepatocellular damage.
