Objective:To investigate the analgesic and anti-inflammatory effect of clematichinenoside(AR),and the therapeutic potential for treating rheumatoid arthritis. Methods:The analgesic effects of AR were observed by acetic acid writhing assay and hot plate in mice. The non-specific anti-inflammatory effect was observed by evaluating xylene-induced ear swelling in mice(acute inflammation)and cotton-induced granuloma in rats(chronic inflammation). The effect on secondary responses of adjuvant arthritis was tested in a rat model of Freund's complete adjuvant arthritis. Results:AR at various doses significantly reduced the number of writhing caused by acetic acid in mice.Especially,AR at high and middle doses significantly prolonged the latency of writhing in mice.AR at various doses significantly improved the pain threshold with different degrees in mice;significantly inhibited ear swelling caused by xylene in mice and cotton-induced proliferation of granulation tissue in rats.AR also significantly suppressed the secondary swelling of Freund's adjuvant arthritis in rats. Conclusion:AR has an anti-inflammatory and analgesic effect, and the effect relates to the dosage.
