Determination of Linoleic Acid in Uygur Medicine Safflower Seed and Safflower Oil by GC

Objective:To develop a GC method with an internal standard for the determination of linoleic acid in Uygur medicine safflower seed and safflower oil.Method:The sample was separated on a TM-WAX(30 m×0.53 mm,1.0μm)column.The purity of nitrogen was 99.99%.The flow rate was 0.5 ml ? min ~ and the split ratio was 5:1.The column temperature was 185℃,the injection temperature was 250℃and the FID detector temperature was 280℃.The flow rate of air and hydrogen was 400 ml·min~(-1) and 60.0 ml ? min~(-1),respectively.Result:The linear range of linoleic acid in safflower seed and safflower oil was 0.199-3.970μg(r =0.999 9, n = 6).The average recovery of linoleic acid in safflower oil was 100.5%with RSD of 1.24%.The average recovery of linoleic acid in safflower seed was 101.9%with RSD of 1.21%.Conclusion:The method is accurate,convenient and reproducible in the determination of linoleic acid in Uygur medicine safflower seed and safflower oil.
