Effect of 4'-Methery-scutellarein on c-myc Expression in Human Choriocarcinoma JAR Cells

Objective To investigate the molecular mechanism of the apoptosis of human choriocarcinoma JAR cell line induced by 4'-methery-scutellarein(4-MS).Methods The techniques of transmission electron microscopy,flow cytometry,real-time quantitative polymerase chain reaction(PCR) and Western blotting were used to study the effect of 4-MS on JAR cells in vitro.Results The morphological changes of apoptosis appeared in JAR cells after treatment with 4-MS.The results of flow cytometry showed that 4-MS induced early apoptosis and cellular necrosis in both concentration-and time-dependent manners,much higher than those in the control group(P 0.01).After the JAR cells were treated by 4-MS at the concentrations of 20 and 40 mg·L-1 for 48 h,c-myc mRNA expression level were 0.87±0.12 and 0.45±0.08,and that of c-myc protein were 0.37±0.02 and 0.25±0.02,respectively.Both the expression levels of c-myc mRNA and c-myc protein were lower than those in the control group(P 0.01).Conclusion 4-MS can induce JAR cell apoptosis,and its mechanism is related to the decrease of c-myc expression.
