Process Research of Decoloration and Deproteinization of Caulis Polygoni Multiflori Polysaccharides

Objective To study the optimal conditions and process parameters of purifying polysaccharides from Caulis Polygoni Multiflori with macroporous resin.Methods With retention rate,decoloration rate and desorption rate of polysaccharides from Caulis Polygoni Multiflori as the observation index,the optimal resin was screened from ten kinds of macroporous adsorptive resins.With retention rate,decoloration rate and deproteinization rate as the index,we optimized the purification process after evaluating the influences of sample concentration,elution flow rate,sample volume and eluent dosage.Results The DA-201 resin was the best of the ten resins,and the optimal purification process conditions were:solution concentration being 1086.6 mg/mL,sample volume being two times as much as resin volume,eluting cubage being 8 times as much as resin volume,distilled water as eluting solvent and eluting rate at 1.0 mL/min.Under the optimal conditions,the retention rate of polysaccharides was 85.42 %,the decolorarion rate was 38.37 %,and the deproteininzation rate was 31.84 %.Conclusion Macroporous adsorption resin is beneficial to removing the protein and pigment of polysaccharides of Caulis Polygoni Multiflori.This method is safe and easy,and can be expected to be used extensively for the preparation of polysaccharides of Caulis Polygoni Multiflori.
