Study on plasma protein binding rate of Rhubarb Free Anthraquinones by equilibrium dialysis method in rats' plasma

Objective: To study the plasma protein binding rate of 5 Rhubarb Free Anthraquinone(FRAs) in rats' plasma.Methods: Under three different concentrations,this paper uses equilibrium dialysis method to imitate the binding process between 5 FRAs and the plasma protein,determines the concentration of 5 FRAs in and out of the dialysis membrane and the binding rate by high performance liquid chromatography(HPLC).Results: Equilibrium dialysis method showed high PBR of these 5 FRAs(80%).Under the experimental concentrations,the PBR of rhein and emodin was inversely proportional to the concentration,and the PBR of physcion varies directly with the concentration.Conclusion: Besides,competitive inhibition between FRAs and plasma protein was revealed,which was considered to be related to the lipid solubility and acidic properties of compounds.
