Evodiamine attenuates acute lung injury induced by lipopolysaccharide in mice

Objective:To determine the protective effect of evodiamine(EVO) on acute lung injury induced by lipopolysaccharide(LPS).Methods:The BABL/C mice were randomly allocated into Normal control group,LPS model group,dexamethasone group(2 mg/kg),EVO group(10 mg/kg),and EVO group(20 mg/kg).The bronchoalveolar lavage fluid(BALF) samples were analyzed for the concentration of numbers of neutrophilic、superoxide dismutase(SOD),methane dicarboxylic aldehyde(MDA) 、interleukin-6(IL-6) and tumor necrosis factor alpha(TNF-α),and The lung samples were taken for histologic evaluation and for determination of wet-to-dry(W/D) lung weight,The protein levels of NF-кBP65 was detected by Western blot analysis.Results:Pretreatment with EVO(10 mg/kg and 20 mg/kg) prior to the administration of intratracheal LPS significantly induced a decrease in lung wet weight/dry weight ratio,in total leukocyte number and neutrophil percent in the BALF,and in MDA activity of lung in dose-dependent manners,and in IL-6,the tumor neurosis factor-α(TNF-α) in the BALF and NF-кBP65 protein in lung.At the same time,pretreatment with EVO also significantly improved the superoxide dismutase(SOD) activity in lung.Conclusion∶ These results showed that anti-inflammatory effects of EVO against the LPS-induced acute lung injury.
