OPTIMIZATION OF MICROWAVE-ASSISTED AQUEOUS TWO-PHASE EXTRACTION PROCESS OF TOTAL FLAVONOIDS FROM MOGHANIA PHILIPPINENSIS

The present study was to examine the optimal conditions of total flavonoids extraction from Moghania philippinensis by microwave-assisted aqueous two-phase,while alcohol and sulfate ammonium solution as the extraction system. Based on the single factor test,the experiment of four factors and three levels was designed with total flavonoids yield as the index according to the principle of central composite design. Furthermore,the response surface methodology was used to analyze the result of experiment and the model of extraction total flavonoids from Moghania philippinensis was established which fits well and truly reflect the relationship between the factors and response values through the verification. The effects of the factors on the total flavonoids yield were determined by regression model. The results showed that the influence order were as follows: the volume fraction of ethanolmicrowave powermicrowave time microwave power: The optimal extraction conditions was:ethanol volume fraction of 41%,addition of ammonium sulfate of 0.28 g·m L-1,microwave time of 21 min and microwave power of 550 W. The predictive value of total flavonoids yield was 6.67%,which was similar to the practical value(6.64%)and the relative error was 0.45%. It verified the stability and reliability of the model.
