Determination of Three Triterpene Acids in Rubi Fructus by Reverse-phase High-performance Liquid Chromatography

Objective To establish a reversed-phase high-performance liquid chromatography(RP-HPLC)method for the determination of three triterpenes(maslinic acid,corosolic acid,and oleanolic acid)in Rubi Fructus,and to provide a basis for the quality assessment of Rubi Fructus.Methods RP-HPLC was performed on a WondaSil C18-WR column(250mm ×4.6mm,5μm)with a mobile phase of acetonitrile(A)-0.2%phosphoric acid solution(B)(9∶1)for isocratic elution at a column temperature of 25 ℃,a flow rate of 0.6mL/min,a detection wavelength of 205 nm,and a sample size of 10μL.Results The concentrations of maslinic acid,corosolic acid,and oleanolic acid showed a good linear relationship with the peak area within the ranges of 0.156-3.900μg,0.102-2.55μg,and 2.04-51μg,respectively,with average recovery rates of95.00%,91.67%,and 93.80%,respectively(relative standard deviations=2.59%,3.25%,and 2.86%,respectively).Conclusion This method is simple and feasible and can be used for the determination of triterpenes in Rubi Fructus.
