The effect of siRNA targeting silencing COX-2 on the expression of VEGF and EGFR in cervical cancer cells

Objective To investigate the effect of siRNA targeting silencing cyclooxygenase-2 gene(COX-2) on the expression of vascular endothelial growth factor(VEGF) and epidermal growth factor receptor(EGFR) of cervical cancer HeLa cells. Method Human cervical cancer cell line HeLa was used as the object of study, HeLa cells transfected by silencing COX-2 gene using siRNA targeting were used as transfection group, HeLa cells without transfection were used as blank control group, HeLa cells transfected by RNA interference gene were used as negative control group. The expressions of COX-2, VEGF and EGFR genes in HeLa cells were detected by reverse transcriptase polymerase chain reaction(RT-PCR); the expressions of COX-2, VEGF and EGFR proteins were detected by Western blot; the proliferations of cells were detected by MTT assay; the apoptosis of HeLa cells was detected by flow cytometry; the changes of migration and invasion ability of HeLa cells were detected by Transwell chamber. Result RT-PCR showed that the expression of COX-2, VEGF and EGFR genes in HeLa cells in transfection group were significantly lower than those in the blank control group and the negative control group(P0.05). Compared with the blank control group and the negative control group,the expressions of COX-2, VEGF and EGFR proteins in the transfection group were significantly lower(P0.05). The results of MTT and flow cytometry showed that the inhibition rate and apoptosis rate of HeLa cells in the transfection group were significantly higher than those of the blank control group and the negative control group(P0.05). Transwell chamber test showed that, compared with the blank control group and the negative control group, the cell penetration rate in transfection group was significantly decreased(P0.05). Conclusion siRNA targeting silencing of COX-2 gene can inhibit the expression of VEGF, EGFR genes and proteins in HeLa cells, further more, promote HeLa cells apoptosis, inhibit HeLa cell proliferation, and reduce the migration and invasion ability of HeLa cells.
