Study on tge effects of aerosolized budesonide on the expression of TGF-β and CTGF in lung tissue of rats with pulmonary fibrosis

Objective:To investigate the effect of aerosolized budesonide(BUD) on the expression of transforming growth factor(TGF)-βand connective tissue growth factor(CTGF)in lung tissue of rats with pulmonary fibrosis.Methods:45 SD rats were randomly divided into 3 groups:normal saline,blank control group(NS group),bleomycin model group(BLM group)and budesonide intervention group(BUD group).After the treatment of 7 d,14 d and 28 d in each group,5 were randomly killed in each group.Hematoxylin eosin(HE)and Masson staining were used to grade lung tissue alveolitis and pulmonary fibrosis degree,and enzyme-linked immunosorbent assay(ELISA)was used to exam the TGF-β and CTGF concentration in bronchoalveolar lavage fluid(BALF).Results:During the course of the experiment,there were no alveolitis and pulmonary fibrosis in group NS group,while alveolitis and pulmonary fibrosis both occurred in group BLM and group BUD(P 0.05).The alveolitis and pulmonary fibrosis of the BLM rats were progressively aggravated.In BUD group,the alveolitis on the 7 th day was the most serious,then began to improve,while the pulmonary fibrosis came worse.The expression levels of TGF-βand CTGF in BALF in NS group rats were lower,and the expression levels of BLM and BUD rats were significantly higher than that of NS group at each time point(P0.05).And the expression level of BUD group was significantly lower than that of BLM group(P0.05). Conclusion:Nebulized budesonide has obvious effect on anti-fibrosis and can significantly decrease the expredsion levels of TGF-β and CTGF in lung tissue of rats.
