Expression of NF-κB and Osteopontin in the Knee Joint Fluid of Patients with Osteoarthritis and Its Clinical Significance

Objective To analyze the expression of inflammatory cytokine kappa B( κB) and osteopontin in knee joint fluid of patients with osteoarthritis and its clinical significance. Methods 46 patients with osteoarthritis treated with total knee arthroplasty were selected as the observation group. 46 patients with traumatic amputation,meniscus injury,cruciate ligament rupture and discoid cartilage injury were treated as control group osteopontin in the knee joint fluid of the observation group and the control group were detected by immunohistochemistry and enzyme-linked immunosorbent assay( ELISA) respectively. According to the guidelines for the diagnosis and treatment of osteoarthritis( 2007 edition),patients with osteoarthritis were divided into early stage,middle stage and late stage. Spearman correlation analysis was used to analyze inflammation molecular nuclear factor κB( NF-κB),osteopontin,osteoarthritis clinical stage,the correlation between inflammatory molecules NF-κB and osteopontin. Results The levels of NF-κB and osteopontin in the knee joint of patients with osteoarthritis were significantly higher than those in the control group( P 0. 05).The expression of NF-κB in the knee joint fluid of patients with osteoarthritis was significantly different among different clinical stages( P 0. 05). The expression of NF-κB and osteopontin increased with the increase of clinical stage. The expression of NF-κB and osteopontin was higher than that of osteopontin( r =0. 714,0. 498,P 0. 05). There was a positive correlation between inflammatory cytokine kappa B and osteopontin( r = 0. 512,P 0. 05). Conclusion The expression of NF-κB and osteopontin in the knee joint of patients with osteoarthritis significantly increase. The osteopontin might be involved in the occurrence and development of osteoarthritis through the inflammatory cytokine κB signal transduction pathway. The level of expression has a positive effect on the severity of the condition.
