Development of Two-site Sandwich Enzyme-linked Immunosorbent Assay with Neuron-specific Enolase Monoclonal Antibody and its Application

An ELISA for neuron-specific enolase (NSE) was developed with 2 NSE-McAbs prepared in our lalyoratory and labeled with HRP. The standard curve was linear and the sensitivity cut-off was 5￣50μg/L. Serum levels of NSE was detected in 23 patients with small cell lung cancer (SCLC) , 32 with non small cell lung cancer(NSCLC) ,22 with other pulmonary disease(BPD) and 36 normal controls(NC). The positivity for NSE was 82. 6% in SCLC (50% in stage Ⅰ and Ⅱ ,85. 7% in stage Ⅲ and 91. 7% in stage Ⅳ),and 3. 1%,4. 5% in NSCLC and BPD,respectively. The serum levels of NSE were significantly higher in SCLC than in NSCLC and BPD(P0. 01) ,and in stage Ⅲ and Ⅳ of SCLC than in stage Ⅰ and Ⅱ(P0. 01). NSE concentrations in partial and complete remission group of 23 patients with SCLC after treatment were significantly lower than those of progression group and non-responsive group. It is concluded that two-site sandwich ELISA with NSE-McAb is highly-sensitive ,specific and easy to perform for the diagnosis of SCLC and also has implication in clinical staging and follow-up of the patients.
