Thermodynamic behavior of doxorubicin hydrochloride interacting with human serum albumin

Purposes—To study the thermodynamic behavior of the interaction between doxorubicin hydrochloride(DOX)and human serum albumin(HSA).Methods—Isothermal titration calorimetry,fluorescence spectroscopy UV spectroscopy and molecular simulation were used to investigate the aforesaid thermodynamic behavior.Result—The binding constants of DOX and HSA were 1.22×10~6,ΔH0,ΔS0;the fluorescence intensity decreased significantly,and the emission wavelength hardly moved.The highest integration of DOX in HSA site I and site II was 9.18 and 8.39,respectively.Conclusion—The interaction between DOX and HSA is electrostatic,and the quenching mechanism is static quenching,and the binding at site I is strong.
