Analysis of screening results by nucleic acid amplification testing in voluntary blood donors in Hangzhou,2011

Objective To investigate the yield situation of HBV,HCV and HIV-1 infection by nucleic acid amplification testing(NAT) in voluntary blood donors of Hangzhou in 2011.Methods The infectious markers of HBV,HCV and HIV-1 in blood samples were screened using PROCLEIX ULTRIOassay and two ELISA kits of different manufacturers.Specimens with an initial reactive on PROCLEIX ULTRIOassay were retested with the ULTRIO assay as well as with the discriminatory reagents.A portion of specimens with only initially reactive on PROCLEIX ULTRIOassay were investigated in the follow-up study.Results Our study showed that 585 samples were ULTRIO initially reactive in the 131 594 blood donations in 2011,with a reactive rate of 0.44%.Among those initially reactive samples,418 could be discriminated as HBV DNA reactive(370 samples,88.5%),HCV RNA reactive(29 samples,6.9%) and HIV-1 RNA reactive(19 samples,4.6%).In the indiscriminated samples,44 were repeated reactive and the other 123 were only initially reactive but failed to discrimination.In all positive samples,there were 97 samples which were judged as HBV DNA reactive but HBsAg non-reactive(NAT yield cases),with a NAT yield rate of 0.07%.A Follow-up analysis was carried out to investigate the 46 specimens which were only initially reactive.The result showed that 6 cases were still ULTRIO initial reactive,with 2 cases which could be discriminated as HBV DNA reactive.Conclusion Our results suggested that the implementation of NAT technique had effectively improved the blood safety and decreased the risk of transfusion-transmitted diseases.The most of the NAT yield cases were HBV DNA reactive while HBsAg non-reactive in the voluntary blood donation population.
