Study on the influence of Chinese population in some of the molecular structure of GPA change on the expression of MN blood group antigen

Objective To investigate the partial GYPA intrion-1,intrion-2 and exon-2 region related to GPA genes in Chinese population,which were related to amino acid changes in the GPA peptide and MN blood type antigen expression.Methods The MN blood group types were tested in unrelated blood donation of Chinese population. The samples were testsd with anti-M,anti-N reaction by serological method. At last,the 150 samples were random selected that had no significant difference between the agglutination intensity about the serological and abstract DNA. The nucleotide sequence of the partial GYPA intrion-1,intrion-2 and exon-2 region were checked by a pair of specific primers. The 150 samples were analyzed the relationship between amino acid in the molecule of GPA changes with M,N blood type antigen specificity expression. Results Through gene sequencing we found that the 4 changed amino acids seat in the GPA chain and this changes did not affect the MN expression of blood group antigen,but the MN genes can be divided into : MGMTMGMG,MTMT,N1N1、N1N2、N2NV,MGN1,MGN2、MGNV、MTN1、MTN2、MTNV. Conclusion Partial changes of the molecular structure of GPA did not affect the expression of MN blood group antigens. This discovery could help to study the relationship between expression of GPA molecular characteristics and MN blood type antigen and lay the foundation for molecular polymorphism tracking Chinese population glycophorin family.
