Comparative study of anti-hepatitis C virus detection using different kinds of assays

Objective To compare the sensitivity and specificity differences between indirect ELISA and sandwich ELISA by testing the same samples. Methods 1 266 blood donation samples and 2 241 clinical samples were screened by two indirect ELISA kits initially,and followed by one sandwich ELISA kit,and then HCV-RNA test. Positive specimens were confirmed by recombinent immunoblot assay( RIBA). Results Comparing with the HCV-RNA test,false positive rate of 2kinds of indirect ELISA and one sandwich ELISA was 0. 37%,0. 23% and 0. 03%,respectively. The positive rate of sandwich ELISA was higher than the two indirect ELISAs,and the analytical sensitivity of sandwich ELISA was more than the indirect ELISA 2 to 6 times. Conclusion The sensitivity and specificity of sandwich ELISA kit was significantly better than those of indirect ELISA kits. If under certain circumstances,the anti-HCV detection method is suggested to be sandwich ELISA and NAT.
