Study on serology and morphology of blood from blood donors with high titer of cold autoanti-Ⅰ

Objective To study serologic features and morphology of red blood cells in additive solution from blood donors with high titer of anti-Ⅰ. Methods Blood group was determined by test tube method and PK7300 instrument. The antibody specificity and titers were detected by test tube method. The exterior change of red blood cell morphology in additive solution was noted. Results Autoanti-Ⅰ and anti-C3 d were detected in the additive solution. The anti-Ⅰ titers were 1∶ 256 at4℃,1∶ 1 at room temperature,and 0 at 37℃. Morphology showed that red blood cells agglutinated at the bottom after mixing with supernatant. Solution became more red,while at the same time,the level of supernatants increased relatively. After shaking several times,the agglutinated red blood cells were mixed with the supernatant. Conclusion Cold agglutinin was sensitive to temperature,leading to disassociation of red blood cells from agglutination. Anti-C3 d was detected on red blood cells,which leads to hemolysis and reddening of supernatant.
