The correlation analysis of the low Jk~a antigen expression and 130A in exon 4 of the JK gene

Objective To analyze the relationship of the low expression of Jk( a + Wb-) blood group phenotype and130 A at Exon 4 of the JK gene in the Chinese population. Methods 3 Jk( a + Wb-) samples were identified from 3800 Shenzhen Han donors via RBC anti-Jka tests and urea hemolysis tests. JK phenotypes of 100 controls were detected by standard serology and urea hemolysis tests. Sequences covering 4—11 exons and partial introns of the JK gene was amplified using genomic DNA by polymerase chain reaction( PCR),and fragments were directly sequenced. Results Among the three Jk( a+Wb-) samples,two carried nt130 G/G in exon 4 and JKB/JKB,the other one carried nt130 G/A and JKA/JKB. The kidd phenotypes of 100 controls were 24 Jk( a+b-),43 Jk( a+b+) and 33 Jk( a-b+). Among the 24 Jk( a+b-) samples,7 individuals carried 6 heterozygous nt130 G/A,17 homozygous nt130 A/A and 1 homozygous nt130 G/G. 7 of the 43 Jk( a +b +)samples carried homozygous nt130 G/G and the rest 36 individuals were heterozygous nt130 G/A. The 33 Jk( a-b+) samples carried nt130 G/G. In addition,homozygous for intron8 84 T/T was found in all 103 samples. Conclusion 130 A in Exon 4 of the JK gene were not associated with Jka+w-expression in this study. The allele is common in the Chinese population.
