Analysis on genetic relationship of 17 Nelumbo nucifera cultivars using AFLP

【Objective】AFLP(Amplified Fragments Length Polymorphism)was used to identify and classify 17 lotus cultivars(including 6 new cultivars and their parents).【Methods】10 primer combinations with high polymorphism were selected from64 AFLP primer combinations using 17 lotus DNA as templates.【Results】A total of 523 fragments were amplified,of which370 were polymorphic fragments,and the polymorphism percentage was 70.75%.Then by cluster analysis,17 lotus cultivars were classified into 2 groups.There were 5 primer combinations,using one of which the 17 cultivars could be separated completely.【Conclusion】AFLP was a reliable and efficient method to identify lotus cultivars.This study provided theoretical reference for classification of lotus and parents selection for hybridization.
