The dynamic expression of hypoxia-inducible factor 1α mRNA in broiler chickens with AS induced by low temperature

【Objective】In order to investigate the relationship between HIF-1αand ascites syndrome,the dynamic expression of HIF-1 a mRNA in hearts and lungs of broiler chickens was measured in the study.【Methods】100 male broiler chickens of commercial strain were randomly allocated to control group(raised at the temperature of(22-24)℃)and low temperature group(raised at the temperature of(9-11)℃)at 15 days old.At 22,29,36 and 43 days of age,six broilers in each group were sampled.Pulmonary arterial pressure(PAP),ascites heart index(AHI)and expression of HIF-1αmRNA in lungs and hearts were determined.And total incidence rates of AS were counted at 15-43 days of age.【Results】The experimental results showed that low ambient temperature induced pulmonary hypertension(higher mPAP),right ventricular hypertrophy(greater AHI)and ascites(22%incidence rate)in broiler chickens.Meanwhile,the expression levels of HIF-1αmRNA in lungs and hearts were significantly increased(P 0.05 or P 0.01)at different time points(22,29,36 and 43 days of age)in low ambient temperature compared with control group.【Conclusions】Our findings hinted that low temperature could increase the expression of HIF-1 alpha mRNA in hearts and lungs of broiler chickens,and HIF-1 alpha might be associated with the phase of development of AS.
