Cloning and sequence analysis of M gene in three porcine epidemic diarrhea virus strains

【Objective】The aim of this study is to investigate the variation of M gene in porcine epidemic diarrhea virus(PEDV)strains emerged in Beijing,Tianjin and Hebei province in recent years.【Methods】The complete M gene of three PEDV strains from several diarrhea samples of piglets from Beijing,Tianjin and Hebei province,respectively,was amplified by RT-PCR.After cloning and sequencing,their sequences were analyzed.【Results】The gene sequences showed that the M gene of all the three PEDV strains was 681 base pair in size,the nucleotide homology among them was above 99.7%,and the three PEDV strains was of one cluster in phylogenetics.Although the three PEDV strains was different from earlier epidemic PEDV strains and the current vaccine strain CV777 in China in phylogenetics of the M gene,the nucleotide homology between them and other selected PEDV strains home and abroad was all above 96.5%.【Conclusion】The M gene of PEDV was still a good candidate for PEDV detection.
