Experimental research of enhancing the sensitivity of cTnI gold immunochromatographic assay by Au nanoparticles

Objective To study a new assay for enhancing the sensitivity of cTnI Glod Immunochromatographic assay quickly by Au nanoparticles. Methods The serum cTnI was qualitatively detected by glod immunochromatographic assay( GICA) and on this basis,the reaction bands of GICA were treated by modification solution,and bound the colloidal gold prepared by NH2OH·HCl reducing HAuCl4,then the diameter of antigen-antibody complex labeled by the colloidal gold was expanded and its color became darker and the signal turned stronger and so the sensitivity was enhanced. At the same time,the intra and between-day repeatability of this method was analyzed. Results The sensitivity was increased 16 times and had better repeatability compared with that of the initial test in this method. Conclusion Colloidal gold prepared by NH2OH·HCl reducing HAuCl4 can be applied as sensitizing reagent under certain circumstances and can enhance the sensitivity of cTnI GICA quickly with stable and reliable result. It may provide a new highly sensitive technical support to the poin-of-care testing.
