Study on the determination method of Ophiopogonin D in Ophiopogonis Radix

Objective To explore the ingredient index of Ophiopogonis Radix and establish a method for determining the index component. Methods HPLC was adopted to determine the content of Ophiopogonin D. The chromatographic column was eighteen alkyl silane bonded silica gel,and acetonitrile-water( 48∶ 52) was used as mobile phase. The detection wavelength was 201 nm and the column temperature was about 25℃. Results Ophiopogonin D sample volume has linear relationship between 0. 06400 and 0. 6400 μg( r =0. 9999); the average recovery rate was 103. 4%( RSD 1. 4%). Conclusion The method is accurate and reliable,which can be used for the determination of Ophiopogonin D and related products in the root tuber of Ophiopogonis Radix and provides a reference for improving the quality control standard of Ophiopogonis Radix.
