Effects of sodium ferulate and atorvastatin on nitric oxide,endothelin-1 and myocardial no-reflow in a rabbit model of acute myocardial infarction and reperfusion

Objective To evaluate the effects of sodium ferulate and atorvastatin on nitric oxide(NO),endothelin-1(ET-1)and myocardial no-reflow in a rabbit model of acute myocardial infarction and reperfusion.Methods Twenty-four rabbits were randomized into 3study groups;8in the control group,8in the sodium ferulate and atorvastatin-treated group(sodium ferulate20mg·kg-1,infusion 5min before operation for 6hours,and atorvastatin 10mg·kg-1,once 2hbefore operation),and 8in the sham-operated group,Animals in the former two groups were subjected to 4hours of coronary occlusion followed by 2hours of reperfusion.Data on haemodynamics were collected.Nitric oxide(NO)and the levels of ET-1in blood sample were determined by radioimmunoassay(RIA).The area of no-reflow was evaluated with pathological means.Results In treated group,the levels of ET-1in blood sample were significantly lower than those in control group(P0.01),and the levels of NO were significantly higher(P0.01).The mean area of no-reflow in treated group(48.54±5.38)% was significantly lower than that in the control group(82.37±4.29)%(P0.01).The area of necrosis area of the treated group(81.25±3.75)% decreased significantly than that of the control group(95.46±4.61)%(P0.01).Conclusion Sodium ferulate and atorvastatin are effective in increasing NO and reducing ET-1in blood plasma,and protecting endothelial cells.It also has a beneficial effect on preventing myocardial no-reflow and myocardial infarction during acute myocardial infarction and reperfusion in rabbits.
