STUDY ON METHYLATION STATUS OF BCL-2 AND P53 GENE IN GALLBLADDER CARCINOMA

Objective:To investigate the potential role of methylation modification(global DNA methylation,bcl-2/p53) in gallbladder carcinogenesis.Methods:Innovative enzyme-linked immunosorbent assay was used to detect global DNA methylation,methylation-specific polymerase chain reaction to detect methylation status of bcl-2 and p53,fluorescent RT-PCR to detect bcl-2 and p53 mRNA expression in 45 cases gallbladder carcinoma and 40 cases chronic cholecystitis.Results:The global DNA methylation status of gallbladder carcinoma was obviously lower than chronic cholecystitis(P0.05).The methylation incidence of p53 in gallbladder was significantly higher than chronic cholecystitis(64.4% vs 17.5%,P0.05);While,there had no significant differences about methylation incidence of bcl-2 between gallbladder carcinoma and chronic cholecystitis(17.8% vs 15.0%,P0.05).bcl-2 mRNA expression in gallbladder carcinoma was obviously higher than chronic cholecystitis,while p53 mRNA expression in gallbladder carcinoma was obviously lower(P0.05).Conclusions:Global DNA hypomethylation and p53 hypermethylation may be an important molecular mechanism in gallbladder carcinoma pathogenesis.
