In Vitro Bacteriostasis and Influences of One Compound and 11 Kinds of Chinese Herbal Medicine on Intestinal Microflora in Turbot

The bacteriostasis influences of extract solutions of one formula YCJ and 11 kinds of Chinese herbal medicines against 11 strains of fish pathogens and on intestinal microflora in turbot Scophthalmus maximus were investigated in vitro by an agar diffusion method and by high-throughout sequencing.The results showed that pomegranate Punica granatum rind and Galla chinensis had the best antibacterial effect,followed by Sculellaria barbata,without obvious antibacterial effect in Patrinia scabiosaefoliaand Lonicera japonica.The extract solution of formula YCJ had different antibacterial effects on different tested pathogens,with better bacteriostatic effects on Vibrio parahaemolyticus,Listonella anguillarum and Staphylococcus aureus,followed by Aeromonas hydrophila and Edwardsiella tarda,and without sigficantly bacteriostatic effect on Enterobacter aerogenes,Proteus mirabilis and Acinetobacter lwoffi.Moreover,the turbot with enteritis fed the mixed herbs powder for 30 days had significantly lower mortality than the fish fed the commercial pellet diet did,with relative protection rate of 47%.There was higher abundance and uniformity of bacterial species in intestine of the fish fed the diets containging Chinese herb medicines,with dominant phyla Proteobacteria and Firmicutes,accounting for more than 90% of all bacterial species.The species proportions of Lactobacillales and Bacillales were significantly increased,while the proportions of Vibrionales,Pseudomonadales and Mycoplasmatales were obviously declined in the intestinal contents in the turbot fed the Chinese herbal medicines.In the intestine tract,however,there was no significant change in the proportion of bacteria except for Mycoplasmatales and Pseudomonadales,significantly declined in the turbot fed the Chinese herbal medicines.The findings indicated that the Chinese herbal formula had a good therapeutic effect on the turbot suffered from enteritis,and a certain influence on the intestinal microflora of turbot.
