The effect of activated microglias on the degree of CREB activation in PC12 cells

Objective To investigate whether activated microglias affected the degree of cAMP response element binding protein(CREB)activation in PC12 cells.Methods Microglias were cultured by primary culture and PC12 cells,by subculture.After being activated by IFN-γ,microglias were co-cultured with PC12 cells by transwell for different periods of time(15 min,2 h and 24 h).After co-culture,Western blot was adopted to detect the total CREB and phosphorylated CREB in PC12 cells, to further determine the degree of CREB activation.Results After 15 min co-culture of activated microglias and PC12 cells,the degree of CREB activation was significantly higher than that of the PC12 cell group(P0.05),which later decreased gradually in a time-dependent manner;at 2 h,no statistical difference could be detected and at 24 h,the degree of CREB activation was even lower than that of the PCI2 cell group.Conclusion Activated microglias can improve the content of activated CREB transiently and rapidly within a short period of time,which helps to intensify the ability of nerve cells in resisting injury.
