Vein transplantation of rat bone marrow-derived mesenchymal stem cells in repair of brain injury in rats

Objective To determine the intracranial expression of grafted cells and the influence on neurological motor function after rat bone marrow-derived mesenchymal stem cells (BMSCs)having been transplanted to Wistar rats with brain injury via the tail vein.Methods BMSCs labeled with bromodeoxyuridine(BrdU)were transplanted via the tail vein to Wistar rats at the first day after brain injury(experimental group 1,n=6),the third day(experimental group 2,n=10)and the seventh day(experimental group 3,n=7).Whole bone marrow pre-labeled with BrdU was transplanted at the third day after brain injury to the rats via the tail vein(experimental group 4,n=6)and alpha-minimum essential medium(α-MEM)was injected to the rats with brain injury via the tail vein(control group, n=6).The neurological motor function of the rats was evaluated at the 1st,3rd,7th,14th day after brain injury,and the intracranial expression of grafted BMSCs was examined by the immunohistochemical staining.Results Scores of neurological motor function evaluation at the fourteenth day after brain injury wereasfollows:26.00±1.00(control group),31.83±1.60(experimental group 1),31.10±1.79 (experimental group 2),28.43±1.72(experimental group 3)and 28.67±1.37(experimental group 4)(P= 0.000).Immunohistochemical staining showed BrdU-positive cells in the injured side of brain tissues of the transplantation groups,which distributed mostly in choroid plexus,and periventricular and perivascular regions.Immunobistochemical double staining of the experimental group 2 found no BrdU+neuron-specific enolase(NSE)and BrdU+glial fibrillary alidic protein(GFAF)double-stained cells.Conclusion Transplantation of rat BMSCs via the tail vein may ameliorate the neurological motor function of rats after brain injury within a certain period of time;the grafted cells have been found to survive in the injured rat brain,but if they may finally differentiate into neurons still await further experimental confirmation.
