Changes of apoptosis-regulating factors in thrombin-induced hippocampal neuron apoptosis

Objective To explore the effect of thrombin(TM)on the expressions of hippocampal neuron apoptosis-regnlating factors,Bcl-2,Bax and c-Jun N-terminal kinase(JNK),and elucidate the possible mechanism ofapoptosis induced by TM.Methods Hippocampal neurons were treated by 40 U/mL TM and stopped to be cultured at different time points(0,6,12,24,48,72 h).Bcl-2 and Bax protein expressions were measured by immuocytochemical techniques;phosphorylated-JNK (P-JNK)and JNK protein expressions were determined by Western blot.Results The expression of Bcl-2 protein of neurons treated by TM was increased at 6 h,as time went by,the Bcl-2 protein expression began to decline at 12 h,to the minimum value at 48 h(P0.01),and began to rebound at 72 h.The expression of Bax protein of neurons treated by TM was increased at 6 h and as time went by, gradually increased until 48 h to its peak(P0.01).The ratio of Bcl-2/Bax was gradually decreased in a time-dependent manner,to the lowest value until 48 h.There was no expression of P-JNK protein at 0 h after TM treatment.P-JNK protein began to be expressed at 6 h,which was significantly different from that of the 0 h group(P0.01),peaked at 12 h(P0.01),lasting until 24 h and gradually declined since 48 h.JNK protein was slightly expressed in the 0 h group;6 h later,its expression was increased(P0.01) and as the acting time of TM prolonged,its expression exhibited no significant changes.Conclusion TM may reduce the ratio of Bcl-2/Bax so as to activate the JNK signaling pathway and induce hippocampal neuron apoptosis through down-regnlating the expression of Bcl-2 and up-regulating the expression of Bax.
