Functional Identification of VDmef2 Gene in Verticillium dahliae Involved in Stress Resistance and Pathogenicity

[Purposes]To identify the function of transcription factor VDmef2 in Verticillium dahliae.[Methods]By virtue of Agrobacterium tumefaciens-mediated transformation(ATMT)method,the VDmef2 gene was knocked out fromV.dahliae.Fungal growth and susceptibility were analyzed to carbon sources and stress conditions,and it also was determined whether VDmef2 gene was required for microsclerotial production on MIM.Cotton(Gossypium hirsutum)seedlings were infected to evaluate the effect of VDmef2 gene knockout on virulence.[Findings]Protein subcellular localization suggested VDmef2 located in the nucleus of V.dahliae.Further assessment revealed that the VDmef2 mutant exhibited a significant delay in microsclerotial formation and was also more sensitive to hydrogen peroxide stress,and showed decreased virulence relative to the wild type strain on cotton.[Conclusions]VDmef2 is essential for microsclerotia formation,resistance against oxidative stress and the pathogenicity of V.dahliae.
