Preliminary Study on Agrobacterium-mediated Transformation into Winter Bud of Mulberry Tree

Mulberry is an important economic tree, which has been exploited commercially in the sericulture industry,agroforestry and horticulture programmers. Traditional breeding methods focused on the hybridization and polyploidy strategies, which were time-consuming, and hardly orientation. Genetic transformation is one of the most reliable and interesting methods for introducing desired genes into mulberry. In this paper, winter bud of variety 72 C002 was chosen to microinject with recombinant Agrobacterium harboring the binary vector pBI121 carrying chimeric β-glucuronidase(GUS) and neomycin phosphotransferase(npt Ⅱ) gene. Expression and presence of transgene was preliminarily confirmed by possible expression of GUS and NPT Ⅱ genes. This result might favorite the innovation and cultivation of mulberry trees in future.
