To screen monoclonal antibodies to progastrin-releasing peptide(ProGRP) and to investigate its significance of monoclonal antibodies to ProGRP in diagnosis of small cell lung carcinoma(SCLC),immunohistochemical method was employed for screening of monoclonal antibodies to ProGRP and detecting the expression of ProGRP in SCLC tissues,NSCLC tissues, benign lung disease tissues.V-B3 of 7 monoclonal antibodies to ProGRP had the highest affinity with ProGRP in SCLC tissues.Immunohistochemical staining results showed that positive expression of ProGRP in 21 SCLC tissues was 90.5% which was located in the cytoplasm or on the cell membrane.Expression of ProGRP could also be detected in 28 NSCLC tissues 28.6% and in 23 benign lung disease tissues 8.7%.There was significant difference in the positive expression rate of ProGRP in 21 SCLC tissues,28 NSCLC tissues and 23 benign lung disease tissues. The conclusion is that detecting the expression of ProGRP in SCLC tissues by immunohistochemical method could be employed to diagnose SCLC disease.
