Vascular development in the segment grafts of Cucurbita in Vitro

The segment of hypocotyl from two species of \%Cucurbita\% were grafted under sterile culture.The vascular development in the graft union from heterograft of \%C.pepo/C.moschata\% and autograft of \%C.moschata\% was examined by using light microscopy.The xylem and sieve elements were differentiated from the parenchyma cells in the scion and the stock,and the callus of the graft interface 6~8 days after grafting.8 days after grafting,the xylem and phloem bridges connecting the scion and stock were formed,and the number of vessels and sieve tubes in the graft interface increased along with the days after grafting.6(5)CF was used as a tracer to test the phloem connection of scion and stock during the different period of development.It was found that 8 days after grafting the tracer could be transported from the scion to the stock.
