Isolation, culture and identification of microvascular endothelial cells from human ovarian carcinoma

Objective To establish a reliable method for isolating and culturing microvascular endothelial cells from human ovarian carcinoma. Methods The tissues of human epithelial ovarian carcinoma were finely minced with scissors, then digested. Mononuclear cells were harvested by Percoll gradient centrifugation. The cells were cultured and identified by immunocytochemical staining and transmission electron microscope. Results Twenty-four hours after inoculation, the cells began to cohere, presented polygonal or fusiform. Eight or ten days later, cells grew as confluent monolayer like cobblestones. Factor Ⅷ related antigen was expressed and W-P body was shown in the cells by transmission electron microscope. Conclusion The microvascular endothelial cells can be obtained in vitro from human ovarian carcinoma with our method.
