Radioprotective effect of genistein on radiation injury in mice

Objective To study the radioprotection of genistein against mice radiation injury.Methods Adult male BALB/c mice were administered orally with genistein at dose of 160 mg/kg 24 h before whole-body γ-ray irradiation.The 30-day survival rate,hemogram of peripheral blood,endogenous hematopoietic spleen colony formation(endoCFUs),and number of nucleated cells,CFU-GM in bone marrow were tested following irradiation.Meanwhile,the radioprotective effect of genistein was compared with that of diethylstilbestrol(DES).Results The 30-day survival rate of irradiated mice subjected to genistein was significantly increased.The endoCFUs,number of bone marrow nucleated cells and CFU-GM in bone marrow in mice pre-treated with genistein were enhanced and endoCFUs number in those mice was 3.47-fold higher than in only irradiated mice.Moreover,the recovery of leukocytes,erythrocytes,lymphocytes and thrombocytes was also observed in mice pre-treated with genistein.The radioprotective effects of genistein were similar to that of DES,but stronger in enhancing survival and promoting recovery of nucleated cells,leukocytes and lymphocytes.Conclusion Genistein was an effective radioprotector.The protective effect of genistein may be related with regeneration of hematopoietic stem cells through not only strengthening radioresistance to reduce radiation-induced damage of hematopoietic cells directly or indirectly,but also enhancing post-irradiation repair or promoting proliferation of hematopoietic stem cells.
